TUDCA was first isolated from bear bile in 1902 and is the major bile acid in bear bile, exhibiting spasmolytic, anticonvulsant, anti-inflammatory, and cholelitholytic activities. Tauroursodeoxycholic acid (tauroursodeoxycholic acid, 1) is the active constituent of bear bile; its chemical name is 2-[[((3α,5β,7β)-3,7-dihydroxy-24-oxocholest-24-en-24-yl]amino]ethanesulfonic acid dihydrate. It was developed by the Italian pharmaceutical company Beside and was first launched in Italy in 1991. In 2007, it was approved for sale in China under the trade name Taurolite and is primarily used in clinical practice for the treatment of cholesterol gallstones, primary sclerosing cholangitis, primary biliary cirrhosis, and chronic hepatitis C. Clinical studies have shown that, compared with ursodeoxycholic acid, compound 1 accelerates stone dissolution, increases the complete dissolution rate, and is associated with no significant adverse reactions.
